Delivery WP2-optimized magnetic particles
- Magnetic Particle silanization (MP800-NH2)
300 µL of a dispersion of MP 800 in PBS (5 mg/mL) and 450 µl of water were added to 1200 µL of ethanol and to 43 µl of APTES (2% v/v solution ). The temperature was kept constant at 50 °C and the reaction was carried out for 48 h.
- Antibody integration onto MP800-NH2 (MP800-AB)
300 µL of 50 mM (1-ethyl-3-(3-dimethylamino) propyl carbodiimide, hydrochloride solution (EDC), 300 µL of N-hydroxysuccinimide (NHS) 50 mM solution and 375 µL of PBS 1x were mixed and left for stirring for 15 min followed by addition of 25 µL of 4 mg/mL fFc antibody fluoresent labeled with A488. The reaction was carried out for 3h at room temperature under continious stirring. After that 1 mL of 100 µg/mL MP 800-NH2 was added and the mixture was left to react overnight at RT. Particles were centrifuged and washed with PBS. Fluorescence spectrum was recorded in PBS 1x at [MP 800 NH2] 100 µg/mL
